Polyclonal programs

Polyclonal antibodies can be generated following our well-established Classical or Speedy 28-Day immunization programs or based on the customer’s SOP’s. Multiple Species are available for immunization from small rodents to large animals. Rabbits, rats, guinea pigs and goats are compatible with our proprietary Speedy 28-Day program. Small rodents are Specific Pathogen Free (SPF). The capacities can be easily adapted to the customer’s requests.

classical to the fastest proprietary program, Speedy 28-Day.

Speedy 28-Day is the reference program to produce custom polyclonal antibodies at Eurogentec.

This proprietary program uses a non-Freund adjuvant that stimulates the immune response and
gives mostly IgG’s, despite the short immunization time.

You receive one shipment:

The pre-immune (negative control), intermediate bleed, and final bleed (unless you decide to adapt the program) are delivered at the end of the program.

Our classical programs provide high titer antibodies with excellent affinity.
Immunization lasts from 65 to 90 days depending on the host and the immune response. We can monitor the immune response after two months using the second intermediate bleed, and deliver the pre-immune and second intermediate bleeds to the customer for on site testing. This allows determining if the final bleed should be maintained as initially scheduled, or if a program adaptation is necessary.

You receive two shipments:

The pre-immune (negative control) and intermediate bleeds, delivered while the program is still running

The final bleed (unless you decide to adapt the program).

The classical programs are recommended when :

Repeating a pre-existing protocol already conducted with a classical program

Immunizing a host where the Speedy 28-Day program is not available
or not guaranteed (chicken, mouse, large animals,…)

Using an antigen of poor immunogenicity that may require a program prolongation

Homology between the antigen and the host

The lower the homology, the higher the immune response.

When working with highly conserved mammalian proteins, it may be useful to use chicken as the host, which produce IgY and not IgG

Antibody batch size expected

If a high batch size is requested, one may immunize one goat or sheep (~1L serum per animal), several rabbits in parallel (~50ml serum each), or immunize one hen and collect several eggs (4 eggs contains as much antibody as the serum of one rabbit)

Application and final assay

If double-staining experiments are to be performed, one may recommend combining one mammal (eg rabbit or mouse) and one hen, because the anti-IgG will not cross-react onto IgY, and vice versa

Quantity of antigen available

Immunizing one mouse requires 15µg protein per injection, while goat immunization will require 200µg per injection

Selection of the best programs depending on the antigen

Anti-antigen

You provide the antigen (peptide, protein, complex sample)

Injecting a complex sample (protein, cell extract…) will result in rich polyclonals, containing a large mixture of different antibodies, some potentially targeting conformational epitopes.

How to provide my antigen

Anti-peptide

Eurogentec designs and synthesizes 15-25 mg of one peptide (≥70% purity, 10-16 aa)

Injecting a peptide will result in a polyclonal containing a limited set of different antibodies, which target sequential -but not conformational- epitopes.

Double X

Eurogentec designs and synthesizes two peptides (15-25 mg each, ≥70% purity, 10-16 aa).

To increase the chances of success the DX programs use two peptides, corresponding to spatially distinct regions of the target protein surface. This increases the opportunity of the resultant polyclonal to recognize the target protein in its natural environment. Each host will receive a combination of the two peptides.

The customer tests the serum from the host(s) and if the assay is positive, we advise that a small purification is performed on each peptide separately. The customer checks the purified antibodies, and determines if one or both peptides did contribute to the signal observed in his assay.

Anti-PTM

Eurogentec designs and synthesizes two peptides: one modified with the PTM (15-20 mg, >70% purity, 10-16 aa) and one unmodified.

The Speedy 28-Day programs are based on frequent boosts and are well adapted to generate anti-PTM Abs as repeated antigen injections over a short period of time may minimize instability of the modification(s) harbored on the antigen peptide.
